Finally tracking down a mint G1 Shockwave a couple years ago did it since I never had this figure as a kid... which sucked because he was always my favorite character.

I also got way more excited than I thought I would when I saw guys like Alternators Hound and the commemorative edition of G1 Soundwave sitting on store shelves. Along with Wheeljack (for some weird reason), those two characters in particular really shoot me back to the beginning of my G1 days as a kid.

Notable mentions go to the first time I saw, bought & held MP Prime in person, and completing my Classics Seekers thanks to the BotCon '07 boxset.

Hmm that would have to be a beat up Hot Rod I got from a convention contact. Long story short. I was not able to get Hot Rod in Montreal and thus went 11 years without never owning one (1986 to 1997, remember ebay was JUST starting). When the seller (who is now a coworker with our toy/comic convention scene in Montreal) was like "Look what I found for you", I screamed like a little girl. It was a major mind f*ck on me.

Most recently, I'd say when I found Alternators Hound, Deadend and Silverstreak at retail. It had been such a long drought between Alternator releases and I skipped like a schoolgirl down the aisle at Walmart.

Three moments come to mind.
Finally finding G1 Punch/Counterpunch in great condition with all weapons.

Finding Alternators Tracks (in original blue box packaging) at a Winners. This was before I knew Alternators were to be re-issued in bubble packaging, and considerably after Canada stopped selling Alternators (skipping Tracks and Meister and stopping ironically at Dead End).

The best "Yes" moment would have to be me armed with a huge amount of disposable income, finding myself in Square One Wal-Mart last summer stepping out of the way of two employees tugging a skid of toys to what looked like McDonalds. Upon closer inspection, the skid was multiple examples of almost the entire last wave of Alternators, on sale for $10 and before the scalpers arrived (or even heard of it). It remains the only time I filled a shopping cart with Transformers, as I bought two of each charactor and barely left a dent in the massive display I was cherry picking from. I doubt I'll ever beat that lucky toy experience.
