I live here, so I can kinda help.. also, depends on which bridge you're using really.
Anyways, when you get into the Falls, look for either Niagara Falls BLVD. or Military Rd. You can hit the Walmart, Target, K-Mart, and Big-Lots here.
Then take the 190 thru Grand Island, take the 290 exit, and get off on the Delaware South exit, take that down until you hit the Target on your right..Keep going and there is a Big Lots a bit further down, and also a Kmart on corner of Hertel and Delaware. One done, back track north up Delaware or any of those roads, and get back on Sheridan/324--take that over until you hit the Niagara Falls BLVD again, where you can go to TRU, follow BLVD north and hit another Target, Best Buy, and Walmart. Get back on Blvd, turn south, get back on the 290, take it until you see the blue water tower and it Y's off..keep left--pay toll, and now you're looking for Lockport/transit Rd exit. That would be Rt78. Exit Left off that after tolls, onto Transit, and a TRU is 1/2 mile down road--continue down a few more miles, hit another Target and Walmart. Turn around, get back on thruway, 90W I believe--but you want to head towards Buffalo, take the Erie/Buffalo/Airport side, keeping left. Take it down to Cheektawaga, get off, the Walden exit--2nd one and go right--a Target and Walmart right there. Get back on Thruway, go south..Get off at Mile Strip Rd/McKinnley Mall exit, there's a TRU in there too. Total drive time, all in all round trip--bout 90mins.
